SV40全名猿猴空泡病毒40（Simian vacuolating virus 40）或猿猴病毒40（Simian virus 40），是一種多瘤病毒，也是一種DNA病毒，有造成腫瘤發生的潛在能力，不過通常會維持於潛伏感染（latent infection）狀態。這種病毒是在1960年，於用來生產脊髓灰質炎疫苗的恆河猴腎臟細胞中發現。

Propagation	Grow cells in T25 ECM-coated flasks(G299) with the following conditions. The base medium for this cell line is Prigrow X medium available in ABM, Cat. No. TM100. To make the complete growth medium, add the following components to the base medium: fetal bovine serum (TM999) to a final concentration of 10% and Penicillin/Streptomycin(G255). Atmosphere: air, 95%; Carbon dioxide (CO2), 5%.
Subculturing	1. Remove and discard culture medium. 2. Add 2.0mL of Trypsin-EDTA solution (TM050) to flask and observe cells under an inverted microscope until cell layer is dispersed. Note: Cells that are difficult to detach may be placed at 37°C to facilitate dispersal. 3. Centrifuge cells at 1500rpm for 3 minutes to pellet. 4. Aspirate out trypsin, leaving pellet undisturbed. 5. Resuspend pellet in fresh culture medium and plate in new culture vessel. 6. Incubate cultures at 37°C . Be sure to use T25 ECM-coated flasks(G299) for growth.
Preservation	1. Freeze Medium: Complete growth medium with 10% DMSO. 2. Storage Temperature: Liquid nitrogen vapour phase.

Abmgood重組hTERT,p53,Myc,Rb,Ras V12, SV40 T antigen於lentivirus vector上, 應用lentivirus的系統, 誘導細胞immortalizing.

DNA Repair Gene Knockdown Cell Lines*

DNA repair proteins maintain the stability of the genome. When repair protein function is impaired through mutations, the genome may become unstable which is a hallmark of solid tumors. The availability of this panel of knockdown cell lines will permit scientists to study the molecular etiology of tumor genomic instability and to exploit it in oncology research. The Knockdown line encompasses the Base Excision Repair pathway, Non Homologous End Joining, and Mismatch Repair pathways.
Trevigen’s Knockdown Cell Lines are target specific LN428 (glioblastoma) shRNA lentivirus transduced cells. They are rigorously qualified and mycoplasma free. The percent knockdown levels range from 63-98% as evaluated by RT-PCR. Lentiviruses are maintained by puromycin selection.
